It's so iconic that people only need to say "Who's On First" in reference to some misunderstanding and everyone knows what it means even if they have never heard the whole skit.
They also often used a theme and variation on it. I have a couple of radio shows where they used similar skits in the same format so it sounded like they changed out the words. That gets old quick but the original lives on.
